Description
The Human von Willebrand Factor (vWF) ELISA quantitates Hu vWF in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu vWF. Principle of the method The Human vWF solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. L’antigène apparenté au facteur VIII, ou facteur de von Willebrand est une glycoprotéine multimérique. Il présente des domaines de liaison fonctionnelle à la glycoprotéine plaquettaire Ib, à la glycoprotéine IIb / IIIa, au collagène et à l’héparine. Le facteur de von Willebrand est synthétisé par les cellules endothéliales et stocké dans les granules de Weibel-Palade. Il médie l’adhérence plaquettaire à la paroi des vaisseaux endommagés et sert de support et de stabilisateur du facteur de coagulation VIII. Le facteur de von Willebrand est l’un des marqueurs les plus utiles pour identifier les lignées de néoplasmes des cellules endothéliales (ou mégacaryocytaires). Comme toutes les cellules endothéliales synthétisent/stockent cette molécule, environ 30 % des tumeurs d’origine vasculaire échouent à la coloration de l’antigène lié au facteur VIII, quelles soient bénignes ou malignes. La coloration de l’antigène lié au facteur VIII a également été utilisée pour mesurer l’angiogenèse, un indicateur de récurrence des tumeurs.
