Description
The Human Uromodulin (UMOD) ELISA quantitates Hu UMOD in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu UMOD. Principle of the method The Human UMOD solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Ce gène code l’uromoduline, la protéine la plus abondante dans l’urine normale. Son excrétion dans l’urine résulte du clivage protéolytique de l’ectodomaine de son homologue ancré dans le glycosylphosphatidylinositol, situé sur la surface cellulaire luminale de la boucle de Henle. L’uromoduline peut agir comme un inhibiteur constitutif de la cristallisation du calcium dans les fluides rénaux. L’excrétion d’uromoduline dans l’urine peut constituer une défense contre les infections des voies urinaires causées par les bactéries uropathogènes. Les défauts de ce gène sont associés à la maladie rénale kystique médullaire 2 (MCKD2) et à la néphropathie hyperuricémique juvénile familiale (FJHN), deux maladies rénales autosomiques dominantes. Ces troubles se caractérisent par une hyperuricémie juvénile, une goutte ainsi qu’une insuffisance rénale progressive. Bien que plusieurs variantes de transcription puissent exister pour ce gène, les natures pleine longueur de seulement deux variantes ont été décrites jusqu’à présent. Elles représentent les principales variantes de ce gène et codent la même isoforme.
